近日,美國(guó)南加州大學(xué)Unmesh Jadhav團(tuán)隊(duì)研究了轉(zhuǎn)錄因子Cdx2在發(fā)育和穩(wěn)態(tài)過(guò)程中如何通過(guò)不同機(jī)制與基因組不同位點(diǎn)結(jié)合,以及這種結(jié)合如何受到DNA甲基化和染色質(zhì)修飾的調(diào)控。研究利用小鼠腸道上皮細(xì)胞作為模型系統(tǒng),通過(guò)多種實(shí)驗(yàn)方法揭示了Cdx2結(jié)合的動(dòng)態(tài)變化及其對(duì)基因表達(dá)的調(diào)控機(jī)制。其中,WGBS(全基因組重亞硫酸鹽測(cè)序)和ChIP-seq(染色質(zhì)免疫沉淀測(cè)序)在本研究中發(fā)揮了關(guān)鍵作用。相關(guān)研究成果以《Motif distribution and DNA methylation underlie distinct Cdx2 binding during development and homeostasis》發(fā)表于Nature子刊《Nature Communications》。
